I'm not a VSE developer, but I would suggest improving the Python API for
the VSE. Currently a lot of actions are only accessible through Operators,
and their parameters and context requirements are very confusing. Also it
would be useful to get access to pixel data of scene frames in preview.

Access to sound samples directly from clips is being worked on from the
Audaspace side. That could be useful for syncing audio recordings, or
scripts detecting beats, combined noise level etc.

I've also been thinking to write a script which optimizes the output GUI
for Video editing. Currently you need the properties tab, which is very
centered around "render me something in 3D", and it could be streamlined to
offer better options for setting up the Video output".
